    SUMMARY OF HUMAN STUDIES ON PNT 200 Bioactive Peptide with Soothing Properties.


    CLINICAL STUDIES on humans were performed by 2 different clinical units in France.


    STUDY NUMBER 1

    • 24 volunteers were included in the study
    • Psychometric tests were used to show the anxiolytic activities of PNT 200 at dose levels equivalent to 2.25 mg and 4.5 mg of peptide per dose twice a day for 15 days prior to testing.

    STUDY NUMBER 2

    • 42 volunteers were included in the study
    • Psychometric tests were used to show the anxiolytic activities of PNT 200 at dose levels of 12 mg of peptide, 2 doses during the 24 hours prior to testing.

    STUDY NUMBER 3

    The main aim of the study was to evaluate anxiety evolution, in a moderate stress situation, after or without 15 days of PNT 200 consumption, by using two appropriate scales:

    1. Cattell Anxiety Scale, providing a general level of anxiety;

    2. Spielberger Anxiety Inventory State-Level including two sub-scales: Anxiety-state scale, a sensitive transient modification indicator for anxiety induced by a stress situation; Anxiety-level scale, evaluating anxiety as a stable disposition.

    Double blind study with 3 parallel groups.

    OPTIMED Statistics Department pre-determined (by randomization) the products given to each subject: Eight subjects (4 males and 4 females) received PNT 200's lowest dose; 8 (4 males and 4 females) received PNT 200's highest dose, and 8 (4 males and 4 females) received the placebo.

    The study was designed with three parallel groups.

    1 group received PNT 200 dose equivalent to 4.5 mg of peptide
    1 group received PNT 200 dose equivalent to 2.25 mg of peptide
    1 group received placebo

    On each test day, subjects arrived at OPTIMED facilities in the morning. Volunteers were asked to eat the product with a standard breakfast. They rested half-an-hour in a supine position before initial blood pressure and heart rate monitoring, performed 3 times within 15 minutes prior to the start of the psychometrics tests, once every 5 minutes.

    Systolic and diastolic blood pressures and heart rate were monitored using an automatic monitor. Measurements were taken one minute after starting each test, and at the end of each test. During all the tests, subject wore an armband on their non-dominating arm that was removed at the end of the test.

    RESULTS

    After the stress test, volunteers with a high anxiety level as a stable disposition had a slower increase on their global anxiety-state score when they received PNT 200 per dose.


    STUDY NUMBER 4

    The main aim of the study was to evaluate anxiety.

    Anxiety was measured with two types of testing during test sessions: The cold pressor test is classified in the provocation type of tests, in comparison with the other types of tests (psychological test like the Stroop test) also used. In practice the volunteers put their hand into cold water (at 2°C), which induces a sympathetic nervous activation, close to an ache, but this reaction is the same kind we observed with other stress causes.

    The volunteers could react differently against the 2 stress tests: physical & psychological. Also 2 different volunteers could react at different levels during the 2 tests. By differentiating the anxiety tests, we would be able to explain or approach the possible reaction mechanism of the peptide. The cold pressor test and the Stroop test are frequently used to check the anxiety activity, also they are perfectly reproductible.

    Randomized double blind study with 2 parallel groups.

    NECKER Hospital Statistics Department pre-determined (by randomization) the products given to each subject: 21 subjects received PNT 200 and 21 received placebo.

    The study was designed with 2 parallel groups.
    • 1 group received PNT 200 equivalent to 12 mg of peptide
    • 1 group received placebo
    Volunteers were asked to take the product with a standard breakfast.

    On each test day, subjects arrived at NECKER Hospital facility in the morning. They rested 90 to 120 minutes in a supine position before initial blood pressure and heart rate monitoring, performed 5 minutes prior to the start of the psychometric and physical tests.

    Hemodynamic Parameters:

    Systolic and diastolic blood pressures and heart rate were monitored using an automatic monitor, Finapres 2300 (Ohmeda - Manepas). Measurements were made 5 minutes after each test.

    Hormonal dosages:

    Cortisol and adrenocorticotropic hormone (ACTH) levels in the blood are stress indicators. Cortisol is a hormone released by the adrenal cortex. ACTH is released by the anterior pituitary gland and acts as a controller of the cortisol secretion. Measurements were made on

    blood samples taken before and after the tests.

    RESULTS

    Hemodynamic Parameters:

    Stroop Test
    During the Stoop test, systolic blood pressure increased significantly lower (p<0.03) in volunteers receiving PNT 200.

    Cold Pressor Test
    During the cold pressor test, the heart rate remained stable in volunteers receiving PNT 200 while it increased significantly (p<0.03) in volunteers receiving the placebo.

    Hormonal Dosages:

    Blood cortisol level remains stable in control group while it decreased significantly (p<0.005) in the volunteers receiving PNT 200.

    During the stress tests, blood ACTH level increases in the control group (p<0.09).

    Conclusion and Recommendations:

    PNT 200 contains a bioactive peptide issued from milk protein hydrolysate, made for human consumption. It has been proven in animal studies as having anxiolytic properties quite similar to those of diazepam. Unlike diazepam, PNT 200 is not habit-forming.

    The recommended dosage for a person 55kg (120lbs) or more is 2 capsules (200 mg each) prior to bedtime. Depending on the individual's stress level, another dose may be taken during the course of the day.
